I'm planning a build with the following components:

Pi 3
HifiBerry Digi+
1 TB USB Hard drive with powered USB port
This will run out to a DAC/amp combo

Can I use Bluetooth to control Runeaudio from my phone?
Does Runeaudio support the official Pi 7" touchscreen?
How do I need to format the USB hard drive (file structure, folder hierarchy) and my music?

No, not at the moment.

Does Runeaudio support the official Pi 7" touchscreen?

Yes, Rune Audio is able to start an optional local browser.

How do I need to format the USB hard drive (file structure, folder hierarchy) and my music?

You can use nearly all filesystems, but i would use a linux filesystem (e.g. EXT4) if it is only used on your Raspberry.
I have my folder stucture like this:
/ARTIST/ALBUM/song

My hard drive will be used by both windows 7 and runeaudio.

I'm planning similar unit. I read you should not have HDD and DAC both on USB. Do you have a concern here?

No, because:
- Ethernet on the Pi also is connected to the internal USB hub
- WiFi is also connected to USB
- may only be an issue with very high sample rates.

I think there is no difference in playing files from a NAS or local HDD to your USB DAC.
I personally prefer I2S DACs, because they are cheap and sound great.
With I2S DACs i never had issues in playing highres files.
On my ODROID i can play files up to 32/384kHz without stuttering.

How do you start an optional local browser on a touch screen

Connect a monitor to your PI2/3 (HDMI or DSI) and it should start automatically.
If it does not check the "local browser" setting on the config page.

The output is optimized for 1024x600 and touch input (no cursor).
