Do not edit the file.
Just enable Spotify in System page. Then, from Spotify app, select runeaudio as target device.

and it seems as though Volumio are also having similar problems with the Merus Amp HAT and the latest kernel drivers, see:

Image files in the 1st post have been updated to latest packages and kernel,
5.4.51. Please try it. If it's still no, we have to wait for the new driver in next kernel.
Anyone with update troubles should try the updated image files as well.
